The driving motivation behind my research is to obtain three-dimensional, high resolution images with quantitative information. The aim is to obtain measurements and parameters, in addition to visual three-dimensional representations, which can inform on the fundamental biophysical laws underlying many biological processes. The main technique I use is cryogenic transmission electron microscopy (cryo-TEM) ideally in combination with light microscopy and other biophysical techniques, and with computer modeling and simulations. I use Single Particle (SP) methods to investigate questions at the molecular and macromolecular level, cryo-electron tomography (cryo-ET) for sub-cellular and whole cell research, and a combination of electron energy loss spectroscopy (EELS) and x-ray energy dispersive spectroscopy (XEDS) to investigate questions related to chemical composition within intact whole cells as well as nanoparticles-bacterial cells associations.
